Steady–State Enzyme Kinetics
[1] Michaelis–Menten Analysis
The intial velocity kinetics of enzyme X were measured. Those data are available here. Plot the data as initial velocity vs. substrate concntration and fit the Michaelis-Menten equation to these data to extract the values of VMAX and KM. Compare the fitted values to the estimated values from the plot. The concentration of enzyme X in each reaction was 125 nM. Calculate kcat and kcat/KM for enzyme X. Is enzyme X a "good" enzyme?
Show Answer
The values of VMAX is 16.6 μmoles•s-1 and the value of KM is 14.0 μM. The Michaelis–Menten plot is shown below. The value of kcat is 133 s-1 and the value of kcat/KM is 9.49 x 106 M-1•s-1. As far as enzymes go, enzyme X is rather good at catalysis under these conditions.
